- What did the researchers consider when
inoculating Morus alba?
When inoculation Morus alba the researchers selected young
growing shoots that contained tips and nodal segments. These shoots were
randomly chosen from 3-year old field-grown trees. The selection and the
sterilisation of the plant material are of utmost importance in order to grow
healthy plantlets. As the exposure time and concentration of the chemical
agents as well as the growth media have a considerable impact on the growth
process these parameters were previously tested. (Aroonpong P., Chang P., 2015)
- Which measures are available for shoot
induction and rooting in vitro?
The induction
of shoots and roots can be influenced and controlled by a specific composition of
nutrients and phytohormons in the growth media. In this study different concentrations
of IBA were tested in MS medium. The authors conclude, however, that
supplementary auxins were unnecessary. All
media contained 3 % sucrose (Sajeevan et al., 2011) and 0.8 % agar. The ph was
also adapted to 5.8. (Aroonpong P., Chang P., 2015)
- Which conclusions do you make when reading
Figure 2 of the publication?
Survival
and endophytic contamination correlate strongly. Shoot tips contain meristem cells
which have not yet been in contact with phloem. Endophytic pathogens have not
succeeded in spreading to this point. The increased
occurrence of browning at nodes 1 and 2 could indicate that young plantlets
lack nutrients. In this situation most energy is stored in the shoot tip and
therefore the young, weak leaves die off first. However, this is a rather vague
interpretation.
The
majority of exophytic contamination at node 3 could be due to the closeness to
the media. At this point pathogens have access to nutrients and hence the best
growth conditions.
- Which factors are crucial for the last
step of in vitro culture: acclimation?
Vulnerable
young plants can react drastically to abrupt environmental changes. It’s
important to acclimatise plants step by step (covering, pots, greenhouse) to
their new surroundings to avoid any kind of shock and negative impacts on
metabolism. Plants need to be well rooted and partially covered when transferred
into pots in order to maintain humidity and temperature. (Aroonpong P., Chang P., 2015)

It is pleasant to read and i have to admit that after reading this review, I've really gained a deeper insight of the content of this scientific research. Your English is clearly above average and there is absolutely nothing to complain about on this topic.
What i would have liked to see is a introduction to the blog post. Just to be remembered why you wrote something in your post. The questions are highlighted and all the references are displayed in a good way. Stay tuned that way. As I wrote above, in term of content I found it revealing and the layout is sharp.

Hi Bettina, you studied the paper in depth indeed. And as far as I see you learnt a lot on this topic. As you write in section three (comment figure 2) some of these interpretations appear to be vague (except of your conclusion on endophytes which did not spread to younger parts). Did you realize explants from node 3 had the lowest survival rate, the highest contamination by endophytes an no browning? Almost no survivors to get brownish leaves... Cheers Hansruedi
